Enhanced production of secretory glycoprotein VSTM1-v2 with mouse IgGκ signal peptide in optimized HEK293F transient transfection

・Mouse IgGκ signal peptide greatly enhanced secretion capacity of VSTM1-v2.
・The yield of VSTM1-v2 in the optimized suspension HEK293F transient expression system was increased by 7 folds.
・VSTM1-v2 was synthesized and secreted as a hyper-glycosylated protein with complex oligosaccharides.
・The purified VSTM1-v2 exhibited excellent bioactivity.
・The study provides a high-efficient way to rapidly enhance production of proteins with weak secretion capacity.

・We described a high-active and stable extracellular β-mannanase produced by a novel mannanase-producer Enterobacter sp. strain N18.
・The mannanase-producer was obtained by high-throughput screening method.
・The β-mannanase is able to decrease the viscosity of guar gum solution in a very short time which implies the potential for serving as a gel-breaker.
・The β-mannanase is characterized by a broad working temperature especially low temperature, salt-tolerance and wide working pH range.

Highlights
・A novel biological 1,4-dioxane (1,4-D) wastewater treatment system was developed.
・The 1,4-D-degrading bacterium Afipia sp. strain D1 was entrapped in gel cubes.
・A high removal rate of 0.67 kg m−3 day−1 and 99% removal efficiency were achieved.
・Significant 1,4-D removal activity was observed even at a low temperature of 6.9°C.
・The activation energy for dioxane degradation was calculated to be 47.3 kJ mol−1.
